The amyloid and tau hypotheses are both inadequate to explain Alzheimer-related pathological change. We intend to use the rich neuropathological data set of NACC alongside datasets from ADNI, ROSMAP, and the Banner cohort in a penultimate synthesis of clinicopathological data. Our goal is to use this data to support an integrative theory of Alzheimer pathogenesis that convincingly captures and explains the spectrum of clinical disease, hoping to unify Alzheimer research behind this model.
